Overdrive is purposely built in and designed to be warm sounding, while increasing the gain will generate stronger distortion and will eventually clip the internal circuits. So if you are dealing with a percussive signal and want to exploit the overdrive, it will take a little experimentation to find the gain setting that sounds good to your ears. Otherwise your optimum gain setting is where the LED occasionally blinks yellow. This is a nice compact indicator of gain while keeping the signal to noise level reasonable.
